Development of Methods for Cytochrome P450 Production
We will develop and optimize methods for high level production of functional, catalytically active human cytochrome P450s by using baculovirus expression in insect cells. Approaches to maximize production of holoenzyme and minimize production of apoprotein will be developed and means to efficiently co-express cytochrome P450:NADPH oxidoreductase will be developed. Human cytochromes P4502A6 and P4502E1 will be used as prototype enzymes for this development. The eventual goal is to develop a complete panel expressing high levels of individual human cytochrome P450 forms. The commercial applications will be for the study of human xenobiotic metabolism.
