Abstract:
THE CONTINUING DEVELOPMENT AND EVALUATION OF NEW RADIOCHELATION AGENTS ARE OF UTMOST IMPORTANCE IN TUMOR IMAGING WITH ANTICANCER ANTIBODIES. WHILE DTPA-LABELED IN-111-RADIOIM-MUNOPHARMACEUTICALS ARE WIDELY USED AS TUMOR IMAGING AGENTS, THEIR HIGH LIVER BACKGROUND IS A DISADVANTAGE IN PROVIDING A HIGH TARGET-TO-BACKGROUND RATIO. THE PROPOSED PROJECT ENVISAGES THE DESIGN AND SYNTHESIS OF SPECIFIC ORGANIC COMPOUNDS WHICH ARE EXPECTED TO BE MORE LIPOPHILIC THAN DTPA AND KNOWN TO COMPLEX RADIOMETALS IN ORDER TO DEVELOP A BETTER CHELATE FOR LABELING ANTIBODIES WITH METALS. EFFORTS WILL BE DIRECTED TOWARD THE SYNTHESIS OF BISTHIOSEMICARBAZONES DERIVED FROM 4-CARBOXYETHYLPHENYL GLYOXAL FOLLOWED BY PURIFICATION AND CHARACTERIZATION OF THE COMPOUNDS. THE NEXT STAGE IS DIRECTED TOWARD THE ANTIBODY CONJUGATION OF CHELATING AGENTS, RADIOLABELING WITH TC-99M AND SEPARATION OF THE DESIRED PRODUCT. THIS WILL BE FOLLOWED BY STUDIES OF THE IN VITRO STABILITY OF THE COMPLEXES TO EVAULATE THE SUITABILITY OF THE CHELATING AGENT, RADIOLABELING WITH TC-99M AND SEPARATION OF THE DESIRED PRODUCT. THIS WILL BE FOLLOWED BY STUDIES OF THE IN VITRO STABILITY OF THE COMPLEXES TO EVALUATE THE SUITABILITY OF THE CHEATING AGENT FOR USE IN THE RADIOIMMUNODETECTION OF CANCER.
