Abstract
A RATE LIMITING FACTOR TO USEFUL GENETIC ENGINEERING IN PLANTS IS THE ISOLATION OF PLANT GENES WHICH CONFER AGRONOMICALLY IMPORTANT PROPERTIES TO CROP PLANTS. TRADITIONAL GENE ISOLATION PROCEDURES WHICH ARE BASED ON A KNOWLEDGE OF THE GENE PRODUCT ARE OF LIMITED USE BECAUSE THEBIOCHEMICAL BASIS OF IMPORTANT PROPERTIES SUCH AS DISEASE RESISTANCE, FERTILITY AND YIELD ARE UNKNOWN. THIS PROPOSAL DESCRIBES THE DEVELOPMENT OF A METHOD FOR THE DIRECT SELECTION OF IMPORTANT GENES IN PLANT CELLS. IT IS A SYSTEMINVOLVING DIRECT "SHOTGUN" CLONING OF PLANT DNA INTO PLANT CELLS COUPLED WITH THE RESCUE OF GENES CONFERRING A PARTICULAR SELECTABLE PHENOTYPE ON THOSE PLANT CELLS. THIS TECHNOLOGY WILL ENABLE THE DIRECT SELECTION AND ISOLATION OFPLANT GENES WHICH CONFER RESISTANCE TO NATURAL TOXINS AND TOXIC CHEMICALS.
