Abstract

WE WILL CONSTRUCT HYBRID POLIO/DENGUE VIRUS GENOMES IN ORDER TO DEVELOP A VACCINE FOR DENGUE VIRUS. IN THE CHIMERIC GENOMES, ONE OF TWO DENGUE VIRUS ENVELOPE GENE FRAGMENTS WILL REPLACE SEGMENT OF THE POLIO VIRUS STRUCTURAL PROTEIN GENES. SUCH CHIMERAE WOULD HAVE THE ABILITY TO REPLICATE IN THE GUT AND TO PRODUCE LARGE QUANTITIES OF IMMUNOGEN WITHOUT INFECTIOUS PARTICLE FORMATION, UNLIKE ATTENUATED LIVE VACCINES OR RECOMBINANT VIRUS VECTORS. PHASE II RESEARCH WILL INCLUDE THE DETERMINATION OF THESE CHIMERIC GENOMES' ABILITY TO ELICIT A PROTECTIVE IMMUNERESPONSE IN MICE, THE CONSTRUCTION AND EVALUATION OF ADDITIONAL CHIMERIC GENOMES, AND THE EXPLORATION INTO PRODUCTION METHODS FOR ENCAPSULATED POLIO/DENGUE GENOMES IN THE ABSENCE OF HELPER VIRUS.
