Abstract
PHASE I RESEARCH WILL EXAMINE THE RELATIONSHIP BETWEEN CYTOTOXICITY, MUTAGENICITY, AND CLASTOGENICITY BY USING A NUMBER OF BIOCHEMICAL TECHNIQUES: DNA ADDUCT FORMATION, DNA/DNA AND DNA/PROTEIN CROSS-LINKING, DNA STRAND BREAKS, AND SEQUENCE ANALYSIS OF DNA. A SELECT GROUP OF CHEMICAL AGENTS WILL BE EVALUATED FOR MUTAGENIC ACTIVITY IN THE MOUSELYMPHOMA (L5178Y/TK+/-) ASSAY SYSTEM BY SELECTING FOR TRIFLUOROTHYMIDINE (TFT) RESISTANCE. SMALL AND LARGE COLONYINDUCTION WILL BE CORRELATED TO CHEMICALLY INDUCED CHROMOSO-MAL ABERRATIONS AND KARYOTYPE ANALYSIS. DNA SEQUENCE ANALYSIS WILL BE PERFORMED WITH BOTH LARGE AND SMALL TK-/- MUTANT COLONIES.
