Abstract
INTERNATIONAL TECHNOLOGIES HAS DEVISED A STRATEGY TO FACILITATE SEQUENCING METHODOLOGY BY USING PHAGE RNA POLYMERASES AND 3'-DEOXYNUCLEOTIDES AND MULTIPURPOSE VECTORS. THIS APPROACH IS EASIER AND FASTER THAN SANGER DIDEOXY SEQUENCING FOR SMALL FRAGMENTS, AS A FRAGMENT ONLY NEEDS TO BE CLONED IN ONE ORIENTATION TO SEQUENCE BOTH STRANDS. PROBLEMS ASSOCIATED WITH SECONDARY STRUCTURE IN SEQUENCING SHOULD BE ELIMINATED WITH THIS TECHNIQUE BECAUSE IT USES DOUBLE-STRANDED DNA AS TEMPLATE. THROUGH THE USE OFMULTIPURPOSE PIBI VECTORS, ONE COULD GO BACK AND FORTH BETWEEN SANGER DIDEOXY AND RNA POLYMERASE SEQUENCING DEPENDING UPON WHICH PROCEDURE GAVE OPTIMAL RESULTS.
