Abstract
GENE (S) CONFERRING TOLERANCE TO THE HERBICIDE DIURON WILL BE ISOLATED BY COMPLEMENTATION OF THE HERBICIDE SENSITIVE WILD TYPE PHENOTYPE OF CHLAMYDOMONAS REINHARDII. DNA WILL BE ISOLATED FROMT HE WELL CHARACTERIZED, DIURON-TOLERANT STRAINS OF CHLAMYDOMONAS (DR8,DR9,AND DR10) AND REPRESENT ATIVE LIBRARIES CONSTRUCTED IN E. COLI. LIBRARY DNA'S WILL THEN BE USED IN TRANSFORMATION EXPERIMENTS IN ORDER TO CONFER DIURON TOLERANCE TO PREVIOUSLY SENSITIVE CHLAMYDOMONAS. THE GENE CONFERRING A TOLERANCE WILL BE RECOVERED FROM TRANSFORMANTS AND CHARACTERIZED. THE GENE WILL BE CONFIRMED TO BE A TRUE TOLERANCE GENE BY RETRANS FORMANTION INTO SENSITIVE CELLS.
